  2. Genetic Mutations May Directly Influence Epigenetic Clocks and Aging Genetic Mutations May Directly Influence Epigenetic Clocks and Aging Scientists have identified a potential link between genetic mutations and epigenetic clocks, shedding new light on the mechanisms behind biological aging. This discovery could reshape how aging is understood and measured. The study explores how DNA mutations accumulating over time may directly influence epigenetic changes, which are often used to estimate biological age. Researchers have examined these connections in detail, aiming to determine whether these processes drive aging or merely reflect it. The findings suggest that a deeper relationship exists between these genetic alterations, which could have significant implications for longevity research. Genetic and Epigenetic Changes in Aging According to a study published in Nature Aging on January 13, a correlation has been observed between random genetic mutations and epigenetic modifications that contribute to biological aging. DNA mutations, which arise from cell replication errors, environmental factors, and the gradual decline of repair mechanisms, have long been associated with age-related diseases such as ******* and neurodegeneration. However, these mutations alone do not fully explain aging. Epigenetic changes, which regulate gene activity without altering the DNA sequence, have been widely studied using “epigenetic clocks.” These clocks track specific DNA markers to estimate biological age. The study suggests that mutations influence these markers, and in turn, epigenetic modifications may affect mutation patterns. This bidirectional relationship has raised new questions about whether epigenetic changes are merely symptoms of aging or active participants in the process. Scientific Perspectives on the Findings Dr. Steven Cummings, Executive Director of the San Francisco Coordinating Center at the University of California, San Francisco, stated to Live Science, that a strong correlation between DNA mutations and epigenetic markers was identified. According to him, the study indicates that mutations at specific DNA sites result in distinct epigenetic changes, leading to cascading effects across the genome. Professor Trey Ideker from the University of California, San Diego, added that a significant loss of DNA methylation was observed at mutated sites, while surrounding areas exhibited increased methylation. This ripple effect extended thousands of base pairs beyond the original mutation, though the precise mechanism remains unclear. He emphasized the need for further research to determine whether mutations trigger epigenetic shifts or vice versa. Implications for Aging Research The study’s findings suggest that genetic and epigenetic changes might be influenced by an underlying process that remains unidentified. Dr. Cummings proposed that DNA mutations could be the primary drivers of aging, while epigenetic changes may reflect this process rather than cause it. If confirmed, this would present a significant challenge for anti-aging research, as reversing mutations is considerably more complex than modifying epigenetic markers. Experts have pointed out that additional studies are required to validate these conclusions, particularly in non-cancerous tissues. The data used in the study were primarily derived from ******* patients, making it necessary to examine whether the same patterns exist in healthy individuals. Longitudinal studies tracking genetic and epigenetic changes over time could provide a clearer picture of their relationship to aging. Further investigations may include laboratory experiments where specific mutations are induced in cells to observe subsequent epigenetic modifications. These insights could help refine the use of epigenetic clocks and lead to a more comprehensive understanding of aging at the molecular level. 4. Gray Zone Warfare Devs Are Testing The Most Requested Features Gray Zone Warfare Devs Are Testing The Most Requested Features Gray Zone Warfare’s developer, MADFINGER Games, continues to innovate and expand Gray Zone Warfare, one of 2024’s most impressive shooters. In April 2024, Gray Zone Warfare emerged to a positive reception, presenting a tactical shooter experience bursting with realism and intensity. It has since sold more than 1.5 million units. Recently, the team at MADFINGER revealed the next lineup of improvements and changes coming to Gray Zone Warfare. The first port of call for these changes is the game’s test server, but if all goes well, they’ll be deployed across the full game soon enough. Gray Zone Warfare is being pieced together with the community in mind, and stacks of feedback have been taken on board and actively considered by the developer. Last year, I was fortunate enough to catch up with MADFINGER’s CEO to ask him about the state of play for Gray Zone and the extraction shooter market in general. While Gray Zone isn’t strictly an extraction shooter, it does have mechanics that align it quite nicely with the genre. Here’s what MADFINGER is testing in the latest test server patch: Introduction of a ‘repair feature’ Easier progression to max vendor level Better disconnect protection Better item and body behaviour and physics The first feature on that list is the most widely requested note in the Gray Zone Warfare community. When implemented, it’ll allow users to patch up their equipment instead of paying a costly price to replace items. As you can expect, items can be repaired, but each successive repair will degrade the overall max quality rating of that item. MADFINGER’s intent to strengthen disconnect protection in the game is also a nice inclusion. Many modern shooters feature this mechanic, and it means that if you disconnect while playing, you’ll load back up with the option to continue where you left off. Gray Zone Warfare has a solid development pipeline lined up, and the passion evident in MADFINGER is a wondrous thing. Have you given Gray Zone a try? Let us know on the Insider Gaming forum. 5. Genesis International: Scottie Scheffler and Rory McIlroy in contention in San Diego Genesis International: Scottie Scheffler and Rory McIlroy in contention in San Diego World number one Scottie Scheffler and Northern Ireland’s Rory McIlroy are in touch with leader Davis Thompson at the halfway stage of the Genesis International in San Diego. American Scheffler hit a five-under 67 to lie one shot behind Thompson, who birdied the final hole as he signed for a 66 to set the pace on eight under. McIlroy also shot a 67, with five birdies in eight holes from the third moving him into contention at Torrey Pines. But the Northern Irishman followed that with bogeys at the 12th and 14th before picking up shots at the 15th and the 17th. He was unable to take advantage of the par-five 18th and is three shots off the lead. “I hit a lot of really good shots. I played well for the most part. I scrambled well,” said the 35-year-old. “I just didn’t take advantage of the par fives. I made up for it in other places, which was nice. But I know going into the weekend I’m going to have to play the par fives better.” Scheffler, who won last year’s Masters, hit only five fairways on Friday and also knows he will have to do better to challenge over the weekend. “I’m definitely going to try to be in the fairway a bit more,” he said. “I felt I survived without driving the ball very well and I’m going to need to drive it better over the last couple of days.” Thompson, 25, claimed his first PGA Tour win last year and had six birdies in his first 13 holes before dropping shots at his next two holes. However, he recovered with a birdie at 16 and then a short-range putt at the last ensured he finished on a high. The Genesis is usually held at Riviera Country Club but was relocated to Torrey Pines because of the Los Angeles wildfires. 6. Twist in the investigation of NSW nurse who went on anti-Israeli rant as morphine vial allegedly found in Bankstown Hospital locker Twist in the investigation of NSW nurse who went on anti-Israeli rant as morphine vial allegedly found in Bankstown Hospital locker There has been a major twist in the investigation of two NSW nurses who went on an anti-Israel rant and claimed they would kill Israeli patients. 7NEWS can exclusively reveal that after Ahmed Rashid Nadir and Sarah Abu Lebdeh were sacked, police allegedly found one vial of morphine in Nadir’s personal locker at Bankstown-Lidcombe Hospital in Sydney’s southwest. Nadir had allegedly asked a former colleague to empty his locker. That staff member reportedly felt uncomfortable with the alleged request and instead called police. That vial was seized and will now form part of the investigation after the shocking video surfaced earlier this week. The video, in which they appear to claim they won’t treat Israeli people and boast of sending them to hell, sparked shock and outrage from other nurses, government officials and the wider community. Camera IconThe two health workers, pictured top, made the comments while talking to Max Veifer on a video chat site. Credit: Supplied On Saturday, police raided Nadir’s house in western Sydney and removed bags of potential evidence. “Officers attached to Strike Force Pearl executed a search warrant at a home in Bankstown about 6pm (on Friday), in connection with an ongoing investigation,” a police statement read. “A number of items were taken for further examination.” Police did not confirm reports they have received a full, unedited video from Israeli influencer Max Veifer, who posted the initial clip. On Friday, NSW Police Commissioner Karen Webb said police wanted the full video to inform investigators considering potential criminal charges. Veifer on Friday shared a longer, two-and-a-half-minute version of his conversation with the nurses in an online chat room. In comments not aired in the shorter, edited version of the video, Veifer asked if his service as an Israeli soldier was why Nadir thought he would go to hell. “Um, that’s definitely the answer, correct,” the nurse replied. The trio then began speaking over the top of each other as they addressed his military service, ****** and the occupied ************ Territories. “One day, your time will come and you will die the most horrible death,” Lebdeh says. Veifer replied: “You spread hate, we spread positivity, we spread protection, we spread peace and you spread death.” Australia’s health practitioner watchdog has updated its public records to show both nurses, who worked at Bankstown-Lidcombe Hospital in Sydney’s southwest, had been forbidden from working in the profession nationwide “in any context”. The pair have also had their registrations suspended by the NSW Nursing and Midwifery Council. CCTV footage has been seized from the hospital and other staff have been interviewed by police. The unfolding scandal has broken trust in the public health system, Premier Chris Minns has conceded, and nurses have also expressed devastation and outrage at the comments. Nadir was treated by emergency services on Thursday night following a “concern for ********”. He has issued an apology through a lawyer after being stood down from the hospital but separately told reporters the incident was a misunderstanding and a mistake before he was admitted to hospital. – With AAP Source link #Twist #investigation #NSW #nurse #antiIsraeli #rant #morphine #vial #allegedly #Bankstown #Hospital #locker Pelican News View the full article at [Hidden Content]

  8. Level Zero: Extraction Updates Canned Two Weeks After Game’s Full Release Level Zero: Extraction Updates Canned Two Weeks After Game’s Full Release Level Zero Extraction has succumbed to one of the issues plaguing the wider industry. The developer behind the title – Doghowl – has taken to social media to announce that no further updates will be rolled out for this extraction shooter that emerged as a 1.0 release just two weeks ago. Level Zero Extraction was shaping up as a tasty, alien-fuelled competitor to Escape from Tarkov, but now, the developer has stressed that the ‘relatively small team’ cannot continue to sustain the game from a financial standpoint. Down the Drain Level Zero Extraction entered early access in August 2024, impressing players worldwide with an innovative take on the extraction shooter genre. In this game, players could assume control of ‘Mercs’ or fearsome aliens, loading into a raid and trying to loot their hearts out before successfully extracting again. Like a few extraction shooters that came before, Level Zero Extraction failed to gain enough traction to sustain the title. At the time of writing, just 10 people are playing the game on Steam, and the title’s all-time peak sits at just 2,601. That’s a paltry condition compared to competing titles like Hunt: Showdown and Dark and Darker, both of which were floating between 10,000 – 12,000 players at the time of publishing. On social media, the Doghowl team wrote: Unfortunately, LZ:E has underperformed compared to our expectations. It’s no longer feasible for our relatively small teams to sustain the game from a financial standpoint. Therefore, it’s a hard but necessary decision to gradually wrap up support. It was stressed that the servers will stay up and wipes will continue to happen, but no further updates will be applied to the game. Now, Doghowl looks to what’s coming next, with the team mentioning that it has plans to develop a new multiplayer title that’s based on a cooperative experience. I want to buy a 2020 Ford truck for $23K in cash, but the dealer is offering $500 off if I get a loan. Is it worth it? I want to buy a 2020 Ford truck for $23,000 in cash — but the dealer is giving me $500 off if I get a loan. If I’m going to pay it off right away with no prepayment penalty, is it worth it? When you have the money to pay for a car in cash, that’s a great way to buy a vehicle. You won’t have to worry about paying interest on a loan and you won’t have a monthly car payment to deal with. However, if you’re being offered a $500 discount for taking out a loan to buy your 2020 Ford truck, even though you have $23,000 in cash to pay for it, it may be tempting to just take on the debt. That could be especially true if you plan to pay it off immediately and there is no prepayment penalty. But, should you follow through with financing the purchase or are there unexpected downsides? When a $500 discount is being dangled in front of you, taking out the car loan would seem to be a no-brainer. If you can read the fine print of the paperwork very carefully to make sure there are no loan origination fees, no prepayment penalties and no hidden costs, passing up the money might seem shortsighted. After all, a car is an expensive purchase and getting $500 off could help make it a little cheaper. Having a mix of different kinds of credit can also be beneficial to you, as it can help your credit score. Getting the loan and paying it off immediately could add a paid-off installment loan to your credit record, which can show lenders that you can manage another type of credit if, say, you only have credit cards otherwise. Again, though, you’d want to do your due diligence and make sure there are absolutely no hidden costs. You’ll also want to be absolutely sure you can trust yourself to pay back the loan in full, right away. Read more: Rich, young Americans are ditching the stormy stock market — here are the alternative assets they’re banking on instead While there are reasons why taking out the loan could make good sense, there could also be some downsides. For one, you may have to fill out a lot of paperwork to get approved for the loan. This can be a hassle, and may not be worth your time. There’s also the impact on your credit score to think about. While having a mix of different kinds of credit can be good, applying for a new loan can also have an adverse impact. Story Continues Specifically, you’ll get an inquiry on your credit record, which typically lowers your score and stays on your credit report for two years. Opening and closing credit accounts — even those paid off in full — can also impact your credit score by shortening the length of your credit history, known as your credit age. Although the negative impact should be temporary, if you are planning for some major borrowing in the months or years after taking the unnecessary car loan — such as buying a home — you may not want to take even the temporary hit to your credit. Lastly, if you aren’t confident that you’ll follow through on loan payback once the car is in your driveway, then you should steer clear. There’s no reason to take the chance of paying potentially costly debt for months or even years just because you were chasing a $500 discount and got sidetracked from using the cash you’d set aside. Ultimately, only you can know if the time spent applying for the credit is worth it, or if you’ll be responsible when borrowing and benefit from the record of a well-managed loan on your credit report. 19. AI to transform wealth management, Microsoft executive says AI to transform wealth management, Microsoft executive says By Oliver Hirt ZURICH (Reuters) – Artificial intelligence will bring major upheaval to wealth management, a Microsoft executive said, as the technology’s potential to process information vastly reduces the hurdles required to compete with established banks. AI’s ability to condense financial data will allow just a few people to offer services that previously occupied entire teams in a bank, said Martin Moeller, head of AI & GenAI for financial services, EMEA, at Microsoft. “Generative AI will reshape the competitive landscape,” Moeller told Reuters. “AI will, for example, significantly lower the threshold for market entry for startups, similar to what the digitalization and internet wave did decades ago.” Since early 2024, Swedish payment service provider Klarna has been using AI from Microsoft’s partner OpenAI which performs the work of 700 employees. The world’s largest asset manager, UBS, also sees great potential for AI. CEO Sergio Ermotti said this month it could boost productivity and make jobs easier. Moeller said generative AI will reduce costs for newcomers, and it can also help family offices, private wealth managers for the super-rich that compete with wealth managers. “Banks that have so far been barely active in wealth management could enter the business with the help of AI without having to invest much in customer advisors,” he said. AI’s advance is gaining momentum from changing customer behaviour, with young entrepreneurs increasingly willing to manage their investments themselves, Moeller said. In response, many banks are working to enable customers to independently consolidate information using AI. “Customers should have access to complex information 24 hours a day, seven days a week,” said Moeller. “Portfolio construction can also be handled by conventional AI.” AI currently does not advise on products or specific investment decisions, but the next stage of development, so-called “agentic AI”, which makes independent decisions without human involvement, is expected to arrive in around two years. His trainer explained the 5 keys to his success. Tom Russell, 40, got in shape while maintaining his busy social and work calendar. He lost 17 pounds by strength training and focusing on eating protein. He didn’t cut out alcohol but drank less to help him lose weight. Tom Russell, a director in the hospitality industry, has a grueling schedule. When he talked to Business Insider from New York City, he was about to head back to London before setting off to Tokyo the week after. He’s also in the best shape of his life. Russell used to work out, but his sessions weren’t structured or efficient. After teaming up two years ago with Harry Cox, a personal trainer who runs London’s private gym ClubQ, Russell dropped 17 pounds over 18 months. He built strength and maintained his busy social life and career — including lots of networking drinks. “I look after teams in Asia as well as right through to the west coast of America. So my day can start relatively early, and then it can run pretty late,” Russell said. He and Cox told *** how focusing on strength training, planning ahead, mindset, and a high-protein diet helped Russell change his approach to health and fitness — and his physique. Russell before working with Cox and 18 months in.Club Q Planning ahead In an average month, Russell spends two to three weeks abroad, he said. This can make sticking to a regular workout schedule challenging. But Cox showed Russell how he could still get results. Every Sunday, Cox checks in with Russell and asks about his plans for the week ahead. Cox then Googles the gyms Russell will be able to access and plans workouts accordingly. “It’s hard enough for him to find the hour a day to get it done, but he doesn’t want to have another hour on top of that figuring out how to do it,” Cox said. Russell has essentially outsourced one element of his life. “He likes the mental freedom of just having to wake up, check his phone, know exactly what he needs to do, and doing it. Then he can get back to all the other things inside his head,” Cox said. Focusing on strength training and steps Russell did strength training to build muscle, lose ****, and improve his health.Club Q For the past two years, Russell has done at least four strength training workouts a week, ideally including one with Cox. Russell’s main motivation was dropping **** and building muscle, so he mainly does bodybuilding-style training, but Cox keeps longevity in mind too. “My responsibility as a trainer is to make sure he doesn’t get injured and his body’s still healthy,” Cox said. So, when Russell is feeling particularly tired or has a more taxing week, Cox doesn’t program exercises like deadlifts which fatigue the body. “Having a high-performing job, one can’t dedicate one’s whole life to the gym,” Cox said. “So it was important to me that he was also able to maintain good energy levels throughout the day. He was not going into the gym and absolutely battering himself.” With little free time, Russell didn’t do cardio workouts, as strength training was a more efficient way to achieve his physique goals. But he tracked his steps and made sure he was walking plenty for his overall health (and calorie burn). Controlling the controllable Entertaining is a big part of Russell’s life, so it’s impossible for him to control or track everything he eats and drinks. With that in mind, Cox encouraged him to “control the controllable” and make smart choices the rest of the time. In practice, that meant lower-calorie, high-protein meals for breakfast (Weetabix, a banana, and a protein shake) and lunch (chicken salad), as well as healthy home-cooked dinners including plenty of protein and vegetables when he was eating in. At events, Russell didn’t worry too much about what he ate and drank but tried to choose lean meats such as chicken breast and fillet steak or white fish. Cutting down on, not quitting, alcohol Russell maintained his healthy lifestyle despite traveling and socializing.Club Q Russell feels working in hospitality comes with certain expectations, including drinking at his company’s events. “We need people to drink alcohol, it’s a huge part of our business,” he said. Russell also enjoys alcohol, so didn’t cut it out. Cox encouraged him to go from drinking five or six evenings a week to two or three. When he was drinking, he stopped after a couple and also prioritized lighter options such as wine over beer. While beer typically contains about 200 calories per serving, a small glass of white wine contains about half that. Understanding the calorie content of alcohol helped Russell realize he wanted to hit his goals more than he wanted to drink a whole bottle of wine. “If I’m going to eat X amount of calories through food, then I’m going to go and lob on another 500 calories through alcohol, then I’m never going to start losing weight,” he said. “And it just kind of put it all into perspective.” Having the right mindset and being disciplined When Russell first approached Cox, he felt ready to make a change. He’d been shocked by his body in vacation photos and decided that instead of feeling sorry for himself, he was going to take action. But Russell knew he didn’t want to do anything drastic or punishing that he could only stick to short-term. “I’m still doing this job, and I’m still being social, I’m still having people over, and we’re still going out,” he said. That said, he has put the work in. “You have to go into it with the right mindset, with discipline,” Russell said. “You say the word ‘discipline’ and everyone sort of shivers and runs away, but discipline doesn’t have to be bad.” “It’s not a negative thing,” he added. “If anything, it actually just gives you some structure so that you know when you want to enjoy yourself and have a good time, you can do it.” Read the original article on Business Insider Source link #busy #40yearold #lost #pounds #drinking #alcohol #traveling #trainer #explained #keys #success Pelican News View the full article at [Hidden Content] For verified travel tips and real support, visit: [Hidden Content]

  24. Mikaela Shiffrin: Skier speaks about ‘visions of crashing’ again after December injury Mikaela Shiffrin: Skier speaks about ‘visions of crashing’ again after December injury The world’s most successful alpine skier Mikaela Shiffrin has spoken openly about her mental health struggles in returning to competition after a freak race injury. The American was set for her 100th World Cup victory when leading the second leg of a giant slalom race in Killington in December when she crashed out near the end of the run, somersaulting into the safety nets. Shiffrin suffered a puncture wound and muscle damage to her stomach, an injury she told BBC’s Ski Sunday was “weird, gross and painful”. The 29-year-old spent two months out injured but is back competing at the Alpine World Ski Championships in Saalbach, Austria. “From the outside you look fine, you’re back skiing again – but you’re not OK yet,” said Shiffrin. “I’ve been experiencing that more than I ever did, or ever imagined, with giant slalom in particular. “Sometimes I’ll get a random vision of crashing. It might not be the Killington ******, it could be the course in front of me, that I have this random vision that I’m in the nets again and something else is stabbing through me.” She partnered with world downhill champion Breezy Johnson in the women’s team combined event on Tuesday, turning down an invite from Lindsey Vonn in the process. The pair won gold. Shiffrin said racing with Johnson had been a different, positive, dynamic, while her fellow American, 29, just wanted to see her happy. The medal was simply a bonus as Shiffrin explained: “As an individual athlete, you never feel you are doing enough. “Be faster, fix other things, there’s always things to work on. You’re living in this world, a very critical world, and you just never feel good enough. “But I could really lean on Breezy. I wanted to do well for her and she felt the same for me.” Shiffrin opted not to compete in the giant slalom but is set for Saturday’s slalom, an event she has won four times. She currently has 15 World Championship medals and would break the record should she finish on the podium again. Shiffrin, however, was unsure of adding to the tally. “It might not be enough for a medal but this is part of the recovery process,” she added.
