The marked zone tends to contain a good amount of Great Mullein almost every round. Screenshot by Destructoid You can also check out any location with the Nature tag, as most plants can spawn within them.

Great Mullein can also occasionally be purchased from Celeste. The item can further be broken down into Assorted Seeds, which is the currency needed to purchase items from Celeste. You can also craft Herbal healing items, Antiseptics, or turn the plants in for the Doctor's Orders quest (also requires Antiseptics, so Great Mulleins can do a 2-in-1 for you).
